TY - THES AB - In preparation for a structure-activity relationship (SAR) study, 11 different analogues of the cytostatic compound cryptophycin-1 were synthesised. Therefore, modified unit C building blocks comprising [alpha]-functionalised [beta]-amino acids were incorporated into the cryptophycin framework. These so-called [beta]2-amino acids were prepared by using conventional Evans' auxiliary chemistry. Furthermore, a novel chiral and cyclic N,N-acetal of benzaldehyde was developed, which may function as a universal [beta]2-amino acid precursor. Finally, "clicktophycin-52" was synthesised, which represents the first cryptophycin triazole analogue.
